Pathological Study on The Antidofal Effects of D-penicillamine in Lead Poisoned Dogs


Abstract
This experiment was undertaken to elucidate pathological change of liver and kidney and hematological change and antidotal effect of D-penicillamine in chronically lead poisoned dogs. Lead acetate were administered to dogs for 12 weeks to
produced
chronic lead poison. To study antidotal effect of D-penicillamine, D-penicillamine(50, 100mg/Kg/day) were administered to dogs for 1 week, after 12weeks' lead treatment.
Lead contents of liver and kidney were significantly increased by chronic lead treatment, and they were decreased by D-penicillamine, however, in whole blood, lead content was increased rather than decreased after D-penicillamine treatment. The
reticulocyte numbers were significantly high compared to control group. But the hemoglobin concentration, Mean Corpuscular Hemoglobin, the number of monocytes and eosinophils of lead poisoned groups were significantly decreased in comparison with
control group. Microscopical section of the liver and kidney of chronic lead poisoned dogs showed histopathological changes. The nuclei in which inclusion bodies were developed, showed very pronounced alterations in size, shape by toluidine blue
staining. It's size were two or three times larger than normal. The cytoplasm of the cells in the liver were swollen and fatty degeneration changes was observed in the cytoplasm.
